Research directions:

  • Manipulation of the recombination process (identification of critical meiotic stages against viral infections, evaluation of the particularities of meiotic crossing-over and the spectrum of aberrations in meiosis in conditions of viral pathogenesis);
  • Variability of quantitative and qualitative traits in offspring derived from parents infected with viruses and / or irradiated with gamma barley (Hordeum vulgare L.) and tomatoes (Solanum lycopersicum L., S. cheesmaniae, S. pimpinellifolium);
  • The cytogenetic specificity of the host-pathogen interaction (histological and ultrastructural aspects, mutational and recombinogenic effects induced in plants, the specificity of the crossingover rate);
  • Application of gametic selection methods in evaluating and identifying the potential resistance of genotypes to environmental factors (viral infections, supraoptimal temperatures, water stress) at the level of male gametophyte in order to streamline the selection process and assess the potential for adaptability.
  • Plant biotechnologies:

    •    induction and evaluation of somaclonal variations generated by tissue cultures of cultivated species (potato, cabbage, wheat, spring barley);
    •    regeneration of haploid plants by in vitro culture of anthers and ovaries (spring barley);
    •    obtaining and multiplying virus-free material (potato, vine);
    •    in vitro rescue technology for immature embryos in the perennial vine varieties;
    •    in vitro micropropagation (Vitis vinifera, Solanum tuberosum, Lavandula angustifolia, Satureja montana, Stevia rebaudiana, Paulownia etc.)

    Scientific skills:

  • Application of in vitro technologies in the creation of valuable forms of different crops, especially cereals (wheat and spring barley).
  • The use of in vitro techniques and experimental mutagenesis based on the application of viruses as a factor for inducing variability and / or gamma radiation, with the possibility of obtaining new forms of crops (tomatoes, barley and wheat) with valuable agronomic characteristics.
  • Obtaining and multiplying virus-free plants. Based on research conducted over 15 years, algorithms have been developed to obtain healthy material for a number of crops - potato, lavender, stevia and vines.
  • Virological diagnosis of vegetative material:
    1. enzyme-linked immunosorbent assay (ELISA)
    2. immunosorbent electron microscopy (ISEM)
    3. electron contrast microscopy with negative contrast

    Relevant results:

    1. It has been established that infections caused by tomato mosaic, tomato aspermia and potato X viruses induce:

    complimented changes in the distribution and exchange rate between homologous chromosomes, significant deviations of mitosis and meiosis processes in host plants.
    2. The assumption regarding the mitogenic effect of phytoviruses was confirmed by the SCE (sister chromatid exchange) test on grain and spring barley.
    3. Experimental mutagenesis methods, based on the use of phytoviruses as inducers of variations, based on different host-pathogen systems, have proven the possibility of expanding genetic diversity, thus obtaining new forms of crop plants (tomatoes, wheat, barley spring), showing positive or negative transgressions.
    4. Based on cytological studies, the restructuring induced by viral infections in the organs of the reproductive system in different crops (tomatoes, wheat, spring barley) was described, being appreciated the reactivity at tissue and cellular level compared to the reorganizations generated by gamma rays.
    5. The optimization of in vitro cultivation conditions and nutrient media allowed the development of working algorithms for micropropagation and ex vitro acclimatization of several plant species of ameliorating interest (Solanum tuberosum, Lavandula angustifolia, Satureja montana, Stevia rebaudiana, Vitis vinifera, Paulownia sp).
    6. In order to obtain the propagating material free of viral infections, the protocols for the eradication of viral twins were prepared by meristem culture with or without thermotherapy.
    7. The application of the in vitro culture technique of immature embryos allowed the elaboration of the procedure for the recovery of the vine seedlings with different degree of apirenia and eradication in this way of the viral germs of the degenerative complex.
    8. For different plant-virus interaction systems (compatible, incompatible, sensitive, tolerant, resistant), transgenerative features of the defensive response of plants were highlighted.
